産品描述 刀豆蛋白(bái)A(Concanavalin A,Con A),來源于Canavalia ensiformis洋刀豆(刀豆),是一(yī)種凝集素蛋白(bái)(Mw 104kDa)。pH≥7.0條件(jiàn)下(xià),Con A以同質四聚體結構形式存在,由四個(gè)分子量為(wèi)26kDa的亞基構成。酸性條件(jiàn)(pH 4.5-5.5)下(xià),Con A解離為(wèi)活化的二聚體結構(52kDa)。每一(yī)個(gè)單體,不管pH值或分子結構如何,都包含2個(gè)金屬離子結合位點。金屬離子(Ca2+ 和 Mn2+ )必須結合到(dào)這兩個(gè)位點上(shàng)Con A才能(néng)發揮活性。乙酰化、琥珀酰化或其他衍生(shēng)物(wù)也能(néng)産生(shēng)穩定的二聚體結構形式。
刀豆蛋白(bái)A(Concanavalin A,Con A)無血型特異性,但選擇性結合發現于各種糖蛋白(bái)、糖脂和糖類分子上(shàng)的α-甘露糖基(α-mannosyl residues)和α-葡糖基殘基(α-glucosyl residues),需要Ca2+ 和 Mn2+ 離子存在才能(néng)顯示活性。Con A适用于碳水(shuǐ)化合物(wù)研究,糖蛋白(bái)純化,酶标記(enzyme tagging),細胞膜研究,細胞凝集,和細胞分型研究。
本品是FITC标記的刀豆蛋白(bái)(FITC labeled Concanavalin A, FITC-ConA),Ex/Em=495/515nm,親和純化所得,基本不含未标記的熒光(guāng)素。建議工(gōng)作濃度範圍是5-20μg/ml。

The animals were then perfused with 100 ml of phosphate-buffered saline (PBS) to wash out blood cells in the vessels, such as erythrocytes and nonadherent leukocytes. After PBS perfusion, the animals were perfused with 25 ml of fluorescein isothiocyanate (FITC)-labeled concanavalin A lectin (ConA; 40 μg/ml in PBS, pH 7.4).
