2. 凝集素的基本介紹
凝集素(lectin)是純化自(zì)各種天然來源(包括種子、植物(wù)根部和樹皮、真菌、細菌、海草(cǎo)和海綿、軟體動物(wù)、魚卵、脊椎動物(wù)和低(dī)等脊椎體液)的糖蛋白(bái)或結合糖的蛋白(bái),因能(néng)凝集紅(hóng)血球(含血型物(wù)質),故稱為(wèi)凝集素。凝集素不具有免疫源性和酶的性質,但具有高(gāo)度專一(yī)的糖結合特異性。凝集素能(néng)夠識别糖蛋白(bái)和糖脂中複雜(zá)的碳水(shuǐ)化合物(wù)結構,也能(néng)識别細胞膜表面的糖基,因此,可作為(wèi)研究細胞膜結構的有利工(gōng)具。另外,凝集素具有多(duō)價結合能(néng)力,能(néng)與熒光(guāng)素、酶、生(shēng)物(wù)素、膠體金等結合而不影響本身生(shēng)物(wù)活性,因此,也常用于免疫細胞化學的研究,以及腫瘤的診斷評價等。
凝集素的體外應用主要有:
1)血型分型和紅(hóng)細胞多(duō)凝集研究(blood grouping and erythrocyte polyagglutination studies);
2)淋巴細胞的促有絲分裂(mitogenic stimulation of lymphocytes);
3)淋巴細胞亞群研究(lymphocyte subpopulation studies);
4)細胞和其他顆粒的分選(fractionation of cells and other particles);
5)正常和病理條件(jiàn)下(xià)的組織化學研究(histochemical studies of normal and pathological conditions);
3. 凝集素的種類和特征
凝集素(lectin)種類繁多(duō),根據不同特征呈現不同的凝集素分類系統,常見(jiàn)的有:
根據凝集素的亞基特征,大緻分為(wèi)四類:部分凝集素;全凝集素;嵌合凝集素;超凝集素;

基本概念:
加納籽凝集素I(Griffonia Simplicifolia Lectin I,GSL I),是分子量約114kDa的一(yī)種糖蛋白(bái)家族,含有兩種亞基,命名為(wèi)A和B,僅分子量稍有不同。這些亞基結合形成四聚體結構,産生(shēng)五種同工(gōng)凝集素(GSI-B4, GSI-AB3, GSI-A2B2, GSI-A3B,GSI-A4 ;或者 BSI-B4, BSI-AB3, BSI-A2B2, BSI-A3B,BSI-A4)。A亞基富集的凝集素傾向于凝集A型血紅(hóng)細胞,但似乎特異性識别α-N-乙酰半乳糖胺(GalNAc)殘基;B亞基傾向于凝集B型血細胞,特異性識别α-半乳糖殘基。商業(yè)化的GSL I是五種同工(gōng)凝集素的混合物(wù),能(néng)結合包括層粘連蛋白(bái)在内的一(yī)些糖蛋白(bái)。
