産品描述 鹽酸二甲雙胍(Metformin Hydrochloride)是最有效和普遍使用的II型糖尿病治療藥物(wù)之一(yī),能(néng)夠降低(dī)肝細胞中高(gāo)血糖症,主要通(tōng)過抑制肝糖原的産生(shēng)(肝髒糖異生(shēng)作用)發揮作用。二甲雙胍(500μM)激活肝細胞AMPK,導緻乙酰輔酶A羧化酶(ACC)活性下(xià)降,誘導脂肪酸氧化,抑制脂肪生(shēng)成酶表達。二甲雙胍(2mM)激活肌肉AMPK,促進葡萄糖攝取。二甲雙胍降低(dī)肝髒葡萄糖生(shēng)成,增加肌肉細胞葡萄糖攝取[1]。二甲雙胍需要肝髒中的LKB1降低(dī)血糖濃度。二甲雙胍(250mg/kg,腹腔注射)以LKB1依賴的方式增加正常飲食小(xiǎo)鼠的肝髒AMPK活性以及降低(dī)高(gāo)脂飲食小(xiǎo)鼠内的血糖水(shuǐ)平超過50%。還(hái)能(néng)減低(dī)ob/ob小(xiǎo)鼠40%的血糖[2]。飲食誘導肥胖症的小(xiǎo)鼠注射二甲雙胍(10mg/kg/天或50mg/kg/天),能(néng)夠降低(dī)體重增加、肝髒脂滴含量,和血漿中總的膽固醇、LDL-膽固醇和甘油三酯水(shuǐ)平[3]。二甲雙胍(250mg/kg)能(néng)夠減少HCT116 p53-\-人結腸癌小(xiǎo)鼠移植瘤模型中的腫瘤生(shēng)長(cháng),但對過表達重組釀酒酵母Ndi NADH脫氫酶的HCT116 p53-\-腫瘤沒效應[4]。
