芝加哥天藍6B(Chicago Sky Blue 6B, CSB6B),又(yòu)稱為(wèi)滂胺天藍(Pontamine sky blue),是一(yī)種染料和谷氨酸鹽衍生(shēng)物(wù),具廣泛的生(shēng)物(wù)活性。芝加哥天藍6B(CSB6B)是囊泡谷氨酸轉運體(vGLUTs)的競争性抑制劑,CSB6B(0.1 mM)抑制大鼠腦(nǎo)部突觸囊泡的谷氨酸攝取(Ki=0.19µM),但不抑制突觸體介導的谷氨酸攝取。CSB6B也是鈣/鈣調素依賴性蛋白(bái)激酶磷酸酶(CaMKP)和其核異構型CaMKP-N 的抑制劑(IC50s分别是4.1和1µM)。CSB6B(17µM)抑制鈉離子-牛磺膽酸共轉運多(duō)肽(NTCP, IC50= 7.1µM),降低(dī)HepaRG細胞感染乙型肝炎病毒(HBV)和丁型肝炎病毒(HDV)。腦(nǎo)室注射CSB6B(0.5µg)抑制小(xiǎo)鼠炎性疼痛,減低(dī)乙酸誘導的扭動和福爾馬林II期的傷害行為(wèi),對自(zì)發運動無影響。CSB6B還(hái)能(néng)被用作複染劑,降低(dī)免疫熒光(guāng)組織化學制備中的背景自(zì)發熒光(guāng)。

配制方法(Preparation):Chicago Sky Blue 6B (CSB) was dissolved in DDW to 10 μM for all in vitro cardiac cell culture experiments, and 50 μM for BMDM experiments.
實驗方法(Assay):Bone marrow cells were isolated and plated in macrophage differentiation medium. After 6 days of culture, the medium was aspirated and replaced with fresh macrophage differentiation medium with the addition of 10 ng/mL LPS. After 24 hours the LPS-containing medium was aspirated, cells were washed twice with PBS, andwere treated with 50 μM CSB in macrophage differentiation medium. Twenty-four hours later the cells were washed and RNA was purified.
體内研究:
動物(wù)模型(Animal Model):12-week-old Hsd:ICR female mice (Envigo RMS)
給藥劑量(Dosages):In vivo: CSB was diluted in PBS. Injection by i.m.: 50 μL of 100 μM stock solution, which is 0.005 μmoles (5 μg). Injections by i.v.: 100 μL of 100 μM stock solution, which is 0.01 μmoles (10 μg).
實驗方法(Assay):Mice were sacrificed 21 days after MI following 7 injections of CSB. The first injection was 50 μL of 100 μM CSB and was injected i.m. into the myocardium. Six additional injections were 100 μL of 100 μM CSB and were injected i.v. into the tail vein. The last i.v. injection was given 14 days after MI. PBS-treated mice were examined for comparison. A total of 6 CSB-treated and 5 PBS-treated mice were examined.

動物(wù)模型(Animal Model):Female C57BL/6 mice (6–10 weeks of age)
藥物(wù)配制(Preparation):1% (w/v) Chicago Sky Blue 6B in PBS
實驗方法(Assay):To visualise implantation sites,mice were injected intravenously with 0.1ml of 1% (w/v) Chicago Sky Blue 6B in PBS under anaesthetic and sacrificed by CO2 asphyxiation 10min later. Individual decidua were removed from the uterus and shaken in 25μl of formamide for 2 days at room temperature (RT), protected from light to extract the dye. After clearing by centrifugation (16,000×g for 5min), dye content in formamide extracts was quantitated spectrophotometrically using an ND1000 Nanodrop at 618nm relative to a standard curve of Chicago Sky Blue dye dissolved in formamide.
